PWT33597

Pathway’s first clinical candidate, PWT33597, is a potent, balanced inhibitor of PI3K alpha and mTOR. There is strong scientific rationale for the utility of targeting both PI3K alpha and mTOR in a broad range of solid tumors. PI3K alpha is one of the most highly mutated oncogenes in human cancers, with frequent mutations or amplifications in a broad spectrum of tumor types, including breast, colon, lung, cervical, ovarian, head and neck and gastric. mTOR, a key integrator of cellular growth and survival signals, is a clinically validated target. Selectively targeting both PI3K alpha and mTOR may represent the optimal approach for knock-down of this important pathway.

PWT33597 demonstrates excellent in vitro selectivity for its targets, with negligible activity against other lipid kinases, protein kinases and other pharmacologically relevant targets at biologically active concentrations. The compound achieves robust pathway inhibition and displays excellent oral efficacy in industry standard xenograft models at well tolerated doses.

Preclinical development of PWT33597 is underway and IND filing is anticipated in Q2 2011. Pathway also has a second generation PI3K/mTOR compound in lead optimization with a differentiated isoform selectivity and pharmacologic profile.



Discovery Programs

Earlier stage discovery efforts are also ongoing around an irreversible PI3K inhibitor and a selective PI3K delta inhibitor. Pathway’s unique chemistry and strong IP estate provide potential to discover and advance novel, differentiated compounds against these important targets.
